Abstract
Breast cancer is a devastating illness, claiming the lives of over 40,000 women each year in the United States alone. As in many diseases, the earlier breast cancer is detected, the more favorable the patient's prognosis. F.or this reason, quantitative staging of disease progression is an important diagnostic goal. At present, only' a qualitative staging system exists. The Furchak lab is engaged in developing an assay for detection of biomarkers of early breast cancer metastasis. Specifically, we are interested in detection of human mammaglobin mRNA, the RNA transcript for a protein that is over-expressed in breast cancer. In the process of developing an assay for the detection of this molecule, the process of producing a full-length human mammaglobin mRNA was explored using ligation to join several smaller oligomers into a full-length DNA template. In the future, mRNA transcripts obtained from this process will be tested in our assay and used to determine the assay's sensitivity to human mammaglobin mRNA.
