Abstract
The Tobacco Etch Virus (TEV), a member of the Potyvirus group of plant viruses, has a (+) sense RNA strand which acts as mRNA inside a host plant cell. TEV RNA codes for a single precursor polypeptide which is autolyticially cleaved into polyfunctional proteins. The nuclear inclusion protein (a) is a replicative and proteolytic protein which accumulates in the nucleus via a known nuclear localization signal (NLS) (Carrington, 1990b). Nuclear localization has been extensively characterized in animal systems, but is largely ambiguous in plants. In this experiment, I have assayed serial deletions from the Nand C termini of the NIa protein NLS for nuclear localization. I have determined the location of the NLS more precisely on the Nla protein and characterized the NLS more extensively.
