Construction and Characterization of Helicobacter pylori tRNA-guanine transglycosylase (TGT) in pET28a His-tagged vector
Abstract
Helicobacter pylorus is a gram-negative, microaerophilic, spiral-shaped bacterium that colonizes the human stomach and infects more than 50% of the human population. Given the high number of clinical cases this bacterium is responsible for, it is a surprise that H. pylorus has yet to be fully characterized. Part of the problem with providing one comprehensive DNA sequence is that H. pylori has a very high mutation rate. TGT is an essential enzyme in the queuine biosynthetic pathway, a pathway that exchanges guanine for the modified nucleoside queuine in the wobble position of tRNAs with anticodon sequence GUN. This pathway serves as a potential target for antibiotic treatment, as it is not found in humans.
