dc.description.abstractIn order to gain a better understanding of the role of the immune system on a molecular level in the development of atherosclerosis, we examined temporal gene expression of 12-lipoxygenase, vascular cell adhesion molecule, and T-cell receptor mRNA in apolipoprotein E knockout mice as well as control C57Bl/6 mice on 1% high fat-high cholesterol diet after 0,1,3,7,14,21,28, and 56 days respectively. Aortic tissue was used as the source of mRNA. mRNA was reverse transcribed and primer pairs for 12-LO, VCAM, and TCR were designed to bind to the complimentary DNA fragments which were then amplified using polymerase chain reaction. We found that VCAM and TCR mRNA was expressed with greater frequency in control than in apo E-/- animals. We conclude that there are no significant differences in expression of these genes between apo E-/- and control mice in the aortic mRNA.en_US
