The Effects of Desipramine on NAAG Hydrolyzing Enzyme, NAALADase

Male Sprague-Dawley rats were injected for 15 consecutive days with either desipramine (25 mg/kg in 0.9% NaCI solution with 10% DMSO) or vehicle (0.9% NaCI solution with 10% DMSO). On the 16th day, the rats were decapitated by guillotine, and the brain was removed and dissected for specific regions. Assays measuring NAALADase activity were then conducted for each region by utilizing NAAG radiolabelled at the glutamate residue. Data from this experiment exhibited a significant difference in the NAALADase activity between DMI and vehicle treated subjects in pyriform cortex. A distinct rostral-caudal NAALADase concentration gradient was also found to be present throughout studied regions of brain.
