Abstract
The objectives of this project were three-fold: (1.) to learn and develop an understanding of disc-gal electrophoresis of lysozyme; (2.) to develop a lysozyme specific staining system using Biebrich scarlet and chitin, and to evaluate its usefulness in disc-gel electrophoresis (Scholnik and Kass, 1973); (3.) to demonstrate isozymes of lysozymle in parotid saljva and other body fluids. Experiments were run using Biebrich scarlet and chitin to stain electrophoretic eels and Zymograms of those gels. Because of the inhibitory interaction of chitin with lysozyme, which left no binding sites for the Biebrich scarlet, this staining technique was judged to be inadequate for further electrophoretic studies. Using the same Zymogram system and a staining system using Swiss blue; isozyme bands were demonstrated in samples of human parotid saliva, serum and the serums of rhesus and green monkeys.
