Effects of Insulin on Cyclic 3'-5- AMP Phosphidiesterase in Relation to Mammary Carcinoma

Insulin has been shown to increase the activity of cyclic AMP phosphodiesterase (PDE) in various rat tissues. It apparently does this by interacting with membrane bound receptors which activate a membrane bound PDE, increasing its activity by raising its Vmax. This membrane bound PDE has been found to be a "low Km" variety of PDE among several other PDE activities. The effect of insulin on POE activity is of importance because of the discovery that cAMP levels have an effect on cell growth, both normal and abnormal. This is of importance in the study of certain hormone-responsive types of cancer. In this study the purpose was to test the effect of insulin on PDE activity on Michigan Cancer Foundation No.7 (MCF7) human mammary cancer cells after first establishing whether or not the cells exhibited multiple PDE activities. The multiple PDE activities were found, but insulin effect was negligible in any at all.
