Study of the Differential Effects of Cytosine Arabinoside on Transformed and Non-Transformed Cells

This investigation was undertaken in an attempt to understand the observed differential growth inhibition of non-.transformed (BALB 3T3) and· transformed (KA31) cells by cytosine arabinoside. The investigation proceeded along two lines. First, the drug effects on these two cell lines in terms of their biochemical and enzymatic aspects, was .studied. Drug effects on DNA synthesis and DNA polymerase were compared. Secondly, the difference, if any, between ·these two cell lines in handling the drug was compared. This included mainly the metabolism of Ara-C, namely deamination, phosphorylation, and incorporation of the drug. Although no differential response to Ara-C was observed between DNA synthesis and cellular DNA polymerase of both cell lines, ·differences in the rate of Ara-C deamination, incorporation, and to a lesser extent, phosphorylation were observed. Differential growth inhibition, between the two cell lines, was also demonstrated.
