Abstract
Cancer accounts for more drug related fatalities in the United States than any other disease related illness, which is why preclinical research and drug development is a popular topic in science today. One of the most important anticancer drug development models is the subcutaneous tumor xenograft model in the nude athymic mouse. The tumor xenograft model was used in this study with various dose regimes of fluorouracil and irinotecan in order to develop a better positive control for future preclinical anticancer drug research. Both drugs were administered alone at different concentrations, and one combination dose was administered with both irinotecan and fluorouracil. lrinotecan was found to be most effective at a dose of 150 mg/kg in decreasing tumor size while also keeping the mice alive and healthy. The combination therapy of 75 mg/kg irinotecan and 50 mglkg of fluorouracil also showed anti-tumor activity and post administration recovery, however, this dose was more toxic than 150 mg/kg of irinotecan alone. In conclusion, irinotecan at a dose of 150 mg/kg was determined to be the best positive control in this experiment. Future studies should incorporate various routes of drug administration, tumor growth models, and other available chemotherapeutic drugs in combination with fluorouracil and irinotecan.
