Abstract
Histone gene expression is coordinated with DNA replication in newly hatched larvae of the brine shrimp Artemia salina. A Charon 4A library or Artemia genomic DNA fragments was screened for histone genes using pPS 101, a 3.4 kb subclone from ~BSh4, an Artemia histone gene recombinant previously isolated from the same library. As a result of this screening two different recombinants were isolated. Upon digestion with the restriction endonuclease EcoR1 one recombinant was determined to be a reconstruction of Charon 4A insert pieces, whereas the second recombinant, ABSh14D, with an insert length of 10.5 kb, gave a digestion pattern different from any previously isolated Artemia histone gene recombinant. Further probing of EcoR1 digested ABSh14D DNA fragments using subclones, pPS 101, pPS 104, pPS 107 and pPS 111 from ~Sh4 gave positive hybridization results for two bands of sizes 7.5 kb and 1.86 kb. Hybridization with pP8 107, a subclone known to contain regions homologous to Drosophila 5S rRNA, also gave a positive hybridization signal for the 7.5 kb fragment indicating there may be regions in ~Sh14D that are, or resemble, 5S rRNA genes.
