An Attempted Tissue Culture Bioassay for Progesterone

A bioassay for progesterone, using accumulation of glycogen in tissue culture as an endpoint, was attempted. To isolate the glycogen from culture a glycogen extraction method using Hyamine Hydroxide was used. This method proved to be too inconsistent to yield a dose-related response. Other methods of extraction were tried but did not yield consistent results when performed on standard glycogen. An enzymatic glycogen assay was used that yields consistently quantitative dose-response curves to standard glycogen solutions.
