Quantitative High Pressure Liquid Chromatographic Analysis of Nanomolar Concentrations of Arabinosyl Nucleosides

At the university of Michigan College of Pharmacy attempts were made to develop a quantitative high pressure liquid chromatographic assay for the antiviral- agent 9~-D arabinofuranosyl adenine (Ara-A) and its metabolite 9-~-D arabinofuranosyl hypoxanthine (Ara-H). Based on a system used to qualitatively separate several adenine related compounds a cation exchange column with microparticulate packing material was used with an isocratic acetate buffer as elucting solvent. Special emphasis was placed on quantifying very small samples~ less than 10 ng of each compound present in the mixture to be separated. Results were mostly negative and expressed the limitations of high pressure liquid chromatographic equipment in quantitative work.
