Abstract
This investigation was undertaken to study the feasibility of using both transformed and non-transformed cell lines as a preliminary screening system for identifying antitumor agents. Agents selected from this testing system should show preferential toxicity towards the highly tumorigenic transformed cells. The three cell lines used in this study were nontransformed BALB 3T3 (A-31) and the transformed cell lines, KA31 and R4 (both subclones of BALB 3T3 cells). The cytotoxicity of several classes of clinically useful anticancer drugs towards these cell lines was determined by both inhibition of cell growth and cell-kill (cloning assay). In the growth inhibition assay only cytosine arabinoside and 6-mercaptopurine showed a marked inhibitory effect of KA31 and R4 cells as compared to the BALB 3T3 cells. Cytosine arabinoside also showed the differential effect towards the two transformed cell lines in the cell-kill assay. None of the other anticancer drugs tested showed this differential cytotoxicity. These results as related to the purpose of this investigation were discussed.
