The In Vitro Effects of Luteinizing Hormone on Rat Ovarian Mitochondria

Mitochondria were isolated from the ovaries of mature rats. Polarographic techniques were used to measure state III resp1ration, State IV respiration, Respiratory Control Ratios (RCR) ,and ADP:O ratios. Addition of Luteinizing Hormone (LH,5O ug) in vitro had no effects on any parameter of ovarian or liver (control) mitochondrial respiration. The effects of 3',5'-Adenosine Monophosphate (c-AMP) and Adenosine Monophosphate (AMP) on ovarian mitochondrial respiration were also studied. Cyclic AMP (0.3 mM) caused significant increases in state III (p<0.00l) and state IV (p<0.0l) respiration, while not affecting RGR or ADP:O ratios. Liver mitochondria, used as a control, were not affected. AMP (0.3 mM) had similar effects as c-AMP. Various methods of isolating ovarian mitochondria were attempted in order to maximize mitochondrial yields while minimizing losses in viability.
