The Study of Erythromycin A Inactivating Enzyme

This study attempted to further investigate the erythromycin A inactivating enzyme obtained from S. coelicolor. First the enzyme was to be isolated and purified. This was to be accomplished by preparing erythromycylamine, an analog of erythromycin with C-9 -> NH2 , and attaching it to an affinity column. Next the enzymatic reaction was to be followed and the products of this reaction isolated. Dihydroerythromycin A-9-3H, which was prepared from NaBT4, was used in order to follow the products of the enzymatic reaction and to see to what extent the dihydroerythromycin A-9-3H was altered by the enzyme.
