In my investigation, I have attempted to: 1) demonstrate that the variety of acceptor amines previously reported to be acetylated by impure preparations of rapid rabbit liver NAT were related to a single species of enzyme not to several isozymes. 2) show the limits on acetylation set by basicity, charge location, and steric effects of various arylamines and to measure the change in Km and Vmax within these series of arylamines, and 3) search for significant correlations between the kinetics of these reactions and the structural characteristics of the acceptor amines such as basicity (pKa), and hydrophobicity.
