Abstract
The antibody dependent cell-mediated cytotoxicity assay (ADCe) has been shown to be more effective in detecting the circulating preformed antibodies involved in hyperacute rejection than the complement dependent serological crossmatch method (CDSC) currently popular in most clinical transplantation programs in the United States. For the time being, ADCC is mainly an experimental technique, which may remain confined to specially equipped laboratories until either it can be modified for use on existing tissue typing apparatus found in hospitals with transplant programs, or the need for its increased sensitivity has been established sufficiently to justify the expenditures involved in equipping a hospital laboratory to run the assay. We have attempted here to replace the 51Cr release readout for the ADCC with a method utilizing a vital dye which would allow the direct reading of results on a phase contrast microscope. This modification would adapt the ADCC for easy clinical use.
