We are interested in locating a single site of action of all or most therapeutically efficacious antidepressants. The new interest in receptor sensitivity has for the most part overlooked the role of antidepressants at presynaptic receptors. Because mianserin has been shown to block Q2-adrenergic receptors26 and since most antidepressants have antiadrenergic effects, 27 we propose investigating the change in a2-adrenergic receptor sensitivity by (3H)-clonidine binding after the chronic administration of a spectrum of antidepressants.
