The Synthesis of Benzo[1,2-b-5,4-b]Ddifuran and Psoralene

The physiological activity of the furocoumarins has raised the question as to the corresponding activity of linear furocoumarons. Unfortunately, phenyl substituted benzofurans exhibit no such activity, but some derivatives of 2-aroylbenzofuran have shown estrogenic activity, and certain bis(chloro-hydroxyalkyl) substituted benzodifurans have shown antimycotic activity. A crude mixture containing 2,6,8-trimethylbenzodifuran has demonstrated a “quite active" erythemal responses. It is this activity that has given the impetus to this present work: the search for a more desirable synthetic route to psoralene and the first total synthesis of a parent linear benzodifuran.
