Binding Immobilization of Phorbol Esters and Diacylglycerols

The goal of this project was to characterize the binding site on protein kinase C (PKC) for tumor promoting phorbol esters. With this information, the hope is that an inert analog of the tumor promoting substances could be made and thus could halt the tumor promotion process. In previous research it has been shown that diacylglycerols (DAG) bind competitively with phorbol esters (PE) to PKC. Because of this and the similar effects that both DAG and PE elicit, it has been proposed that these two compounds are endogenous analogs. The thrust of this project is to obtain direct proof of this by gaining physical evidence that both bind to the same site. This is done using spin and fluorescent labels to look at binding immobilization. This project involves preparing fluorescently and spin labeled DAG’s and a fluorescently labeled PE. The binding affinity of the DAG’s are then characterized and the binding immobilization studies are done.
